1:稀土摻雜上轉換氟化物納米顆粒在生物分子檢測中的應用
利用靜電吸引層層組裝的方法在NaYF4BYb3+,Er3+外部引入氨基,從而能夠和生物素相連,隨后體系中再加入同生物素相連的金納米顆粒,當抗生物素蛋白存在的時候會連接兩種納米顆粒,從而發生能量轉移,此時金會吸收稀土上轉換納米材料的熒光,通過熒光猝滅的程度實現抗生物素蛋白的檢測,檢測范圍為0.5~370mol/L。

米顆粒結合磁場生物分離技術用于微量DNA的檢測。非常巧妙的利用Lemieux2vonRudloff試劑直接氧化合成過程中引入的油酸配體,得到羧基修飾的上轉換納米顆粒,用于定量檢測靶向DNA。將Fe3O4和NaYF4BYb,Er納米顆粒包覆在二氧化硅微球內,制備的發光磁性多功能復合納米顆粒,在生物分離、檢測、標記成像及藥物輸送方面有著潛在的應用價值。
2:稀土摻雜上轉換氟化物納米在免疫分析中的應用
2001年,Niedbala課題組報道了上轉換納米顆粒應用于免疫層析實驗。研究者將上轉換材料同生物分子相連后,采用免疫層析技術對抗原進行檢測取得了理想的結果。由于層析所用的底板在紅外光照射下不發光,所以觀測到的信號必然來自上轉換納米顆粒,因此上轉換納米材料用于免疫層析技術更加提高了這種方法的可靠性。400nm的UCNP與Cy5相比,目標物濃度和發光強度的線性關系提高兩個數量級。此外,研究者將膠體金和上轉換納米顆粒同時應用于HPV16的檢測,結果表明UCNP實驗的檢測限低于膠體金的100倍。但需要說明的是,控制上轉換納米顆粒的尺寸是擴展其在免疫層析技術應用的關鍵因素。
3:稀土摻雜上轉換氟化物納米顆粒在生物標記中的應用
稀土上轉換氟化物納米顆粒在生物標記中的應用成為當前納米標記的熱點。初的活體實驗是用線蟲吞噬稀土上轉換氧化物納米顆粒。制備了氨基修飾的摻雜FITC的上轉換二氧化硅納米顆粒,又將能與癌細胞特異性結合的葉酸共價鍵連接在納米顆粒外面。這種納米顆??梢噪p模式上轉換下轉換發光,同時實現對癌細胞的標記,用于流式細胞儀進行分選,將葉酸修飾到聚乙烯亞胺包覆的NaYF4BYb,Er納米顆粒上,進行了一系列的細胞以及白鼠活體實驗。研究表明此納米顆粒在磷酸鹽緩沖溶液中非常穩定,同時對骨髓干細胞沒有毒性,抗漂白性好。將其應用于人卵巢癌細胞和結腸癌細胞細胞標記,修飾后的納米顆粒和細胞表面具有較高的親和力。小白鼠實驗中,將UCNP和量子點同時應用于皮下組織成像,在紫外光照射下腹部皮下組織的量子點沒有光信號,而在980nm的激發下,可以清楚看到UCNP的發射光,證實了UCNP在生物活體標記中的潛在價值。
4:稀土摻雜上轉換氟化物納米在光動力治療中的應用
次將稀土上轉換納米顆粒作為光敏劑的能量供體,包覆二氧化硅的同時摻雜部花青,實現UCNP和部花青的能量轉移產生單線態氧。將約50nm大小的共價連接葉酸、非共價吸附酞菁鋅的聚乙烯亞胺包覆的NaYF4BYb,Er納米顆粒,應用于活體實驗。酞箐鋅的吸收波長和NaYF4:Yb,Er的670nm處的發射有重疊,發生能量轉移,當用980nm的激發上轉換納米顆粒時,光敏劑會產生單線態氧。將這種多功能的納米顆粒應用于活體實驗,證明了此種納米顆粒不僅細胞毒性小,而且可以靶向識別癌細胞,并且可以有效的殺死腫瘤細胞,癌細胞殺死率高達80%。
